糖原染色是病理學中常規(guī)的染色方法之一，McManus在1946年最先使用高碘酸-雪夫技術顯示黏蛋白，該法常用來顯示糖原和其他多糖，該技術不僅能顯示糖原，還能顯示中性黏液性物質和某些酸性物質以及軟骨、垂體、霉菌、真菌、色素、淀粉樣物質、基底膜等。過碘酸(又稱高碘酸)是一種強氧化劑，它能氧化糖類及有關物質中的1，2-乙二醇基，使之變?yōu)槎?，醛與Schiff試劑能結合成一種品紅化合物，產生紫紅色。由于高碘酸還可氧化細胞內其他物質，使用時應注意選擇好高碘酸濃度和氧化時間，使氧化控制在既能把乙二醇基氧化成醛基又不至于過氧化，這是很關鍵的步驟。
